
HUT-78 cells (T, Targets, 5 x 103 cells/200μL/well) were incubated with increasing concentrations of anti-CD95 overnight in a 96-well plate. At the last 4 hours of the incubation, 10μM BrdU was added. The cells were then processed using the Ziva®-Tox Cytotoxicity Assay. Specific killing was calculated using the following equation:
% Specific Killing = { 1 - [(RLUexp-RLUspont) / (RLUmax-RLUspont)] } x 100%Where RLUexp are the relative light units from the experimental wells, RLUspont represents the background signal from unlabeled HUT-78 and RLUmax are the relative light units from the untreated HUT-78 cells. RLUspont did not exceed 5% of the RLUmax. Error bars represent Standard Deviation of the mean of triplicates. As a negative control cell line, K562 human CML cells were treated in parallel.
