Ziva®-Tox Is Compatible with the Standard 4-hr Cytotoxicity Incubation
Left Panel: NK92 cells were harvested 7 days post IL-2 stimulation and incubated with K562 cells (5x103 cells per well).
Right Panel: Naïve mouse splenocytes were incubated with YAC-1 cells (5x103 cells per well).
In each experiment, the cells were incubated at the indicated E:T ratios for 4 hrs in a CO2 incubator. During the last 3 hours, BrdU was added to label proliferating cells. Bars represent specific cytotoxicity calculated from triplicate wells ± standard deviation.
Spontaneous BrdU incorporation was defined as the amount of BrdU incorporated by cells in the presence of 0.1% azide. These wells did not exceed an average value of 9% of the maximum incorporation. The highest spontaneous incorporation values occurred at the 10:1 (16%) and 100:1 (25%) ratios.
